| Tumorigenic: | No |
| Effects: | No, did not form colonies in soft agar |
| Comments: | The cells lack B- and T-cell surface markers including T-associated antigens, E-rosetting capacity, surface and intracytoplasmic immunoglobulins. They are non-specific esterase positive. The cells are phagocytic for latex beads and iron particles. Exposure to phorbol 12-myristate 13-acetate (TPA) increases the phagocytic activity. TPA also induces macrophage-like differentiation. The cells are negative for Epstein-Barr virus nuclear antigen (EBNA-). |
| Complete Growth Medium: | The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, ATCC 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (ATCC 30-2020) to a final concentration of 10%.
|
| Subculturing: | Cultures can be maintained by addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension in fresh medium at 3 - 5 x 10 5 viable cells/mL. Maintain cultures at cell concentrations between 3 x 10 5 and 3 x 106 viable cells/mL. Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density). |
| Cryopreservation: | Freeze medium: Complete growth medium 95%; DMSO, 5% Storage temperature: liquid nitrogen vapor temperature |
| Culture Conditions: | Temperature: 37°C
|
| Population Doubling Time: | 24 to 36 hrs |
