| Applications: | The cell line was established by Louis Gastinel and Pamela Bjorkman in 1992. Spleen cells were fused with HL-1 Friendly myeloma cells (a derivative of P3X63Ag8.653 that is deficient in both hypoxanthine phosphoribosyl transferase HPRT and adenine phosphoribosyl transferase APRT). FcRn is a heterodimer composed of a membrane bound heavy chain attached noncovalently to beta 2-microgloublin. This antibody is used in studies of functions of the MHC class I light chain. |
| Storage Conditions: | liquid nitrogen vapor phase |
| Derivation: | The cell line was established by Louis Gastinel and Pamela Bjorkman in 1992. Animals were immunized with and twice boosted with soluble neonatal Fc receptor (FcRn) in an adjuvant. Three days before fusion one mouse was boosted with purified FcRn. Spleen cells were fused with HL-1 Friendly myeloma cells (a derivative of P3X63Ag8.653 that is deficient in both hypoxanthine phosphoribosyl transferase HPRT and adenine phosphoribosyl transferase APRT). |
| Genes Expressed: | immunoglobulin; monoclonal antibody; against rat beta2 microglobulin |
| Cellular Products: | immunoglobulin; monoclonal antibody; against rat beta2 microglobulin |
| Comments: | The cell line was established by Louis Gastinel and Pamela Bjorkman in 1992. The antibody recognizes rat beta 2-microglobulin alone or complexed with FcRn heavy chains. FcRn is a heterodimer composed of a membrane bound heavy chain attached noncovalently to beta 2-microgloublin. It is structurally similar to class I major histocompatibility (MHC) molecules. This antibody is used in studies of functions of the MHC class I light chain. |
| Complete Growth Medium: | HL-1 medium supplemented with 4 mM L-glutamine, 1 mM sodium pyruvate and 1% fetal bovine serum. HL-1 medium can be obtained from from Lonza (catalog number 77201).

| Subculturing: | Subcultures are prepared by scraping. Dislodge cells from the flask substrate with a cell scraper. Aspirate gently and dispense into new flasks. Seed flasks at 2xa0x 105 viable cells/mL. Subculture Ratio:xa01:5 every 3xa0to 4 days. Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density |
