| Comments: | Add contents of thawed ampule to a 80-85% monolayer of HDF cells. If foci of CPE fail to spread by day 11 PI, split infected cells 1:2 and observe for CPE as monolayer reforms. Attenuated for man, broad antigenicity. Stable for 6 months at -60°C in form of glycerinated cells. Virus primarily cell associated. A long (10-11 day) incubation is required for initial passage of frozen stocks. |

Recommendations For Infection: Trypsinize a 2-4 day old cell culture and seed at a density of 105 cells per cm2 (2.5X106 cells/T25 flask; 7.5X 105 cells per T75 flask) using the virus growth medium (VGM). To the freshly trypsinized cells inoculate using a MOI of 0.1. Immediately add additional VGM depending upon the size of the flask used (recommended medium volume for T25 is 5 to 7 mL and for T75 15 to 22.5 mL). Incubate for 24 hours at 37°C in a 5% CO2 atmosphere. Aspirate media and add fresh virus/cell growth medium to remove any traces of DMSO. Continue incubation.
Incubation: 2-7 days in a 5% CO2 air atmosphere in a stationary position until CPE is seen
