| Comments: | Protoarchetype regulatory region; naturally severely attenuated from microRNA expression. SV40 regulatory region, nucleotides 5192-370; E = enhancer region within regulatory region; 1E = single 72-bp enhancer (archetypal); 2E = duplication/rearrangement in enhancer region (complex), nucleotides 107-294; G/C-rich region within regulatory region, nucleotides 40-103. Nucleotide numbers from SV40 reference strain 776-2E. |
| Effect on Host: | CPE, vacuolization, rounding, and sloughing |
| Recommended Host: | CV-1 cells (ATCC® CCL-70™) |
| Growth Conditions: | Temperature: 37°C Recommendations for Infection: Plate cells 24-48 hours prior to infection and infect when cultures are 80-90% confluent. Remove medium and inoculate with a small volume of virus (e.g. 1 mL per 25 cm2) diluted to provide an optimal MOI (e.g. 0.01). Adsorb 2 hours at 37°C in a humidified 5% CO2 atmosphere. End adsorption by adding virus growth medium. Incubation: 9-16 days at 37°C in a humidified 5% CO2 atmosphere, until CPE is progressed through 80% of the monolayer. |
