Karyotype: The reported karyoype was 46, XY, dup(1) (pter to q32::q21 to qter), t(8;14) (q24.1; q32), -10, +der(10)t(Y;10)(q12:q26), -21, +der (21) t (?; 21) (?; p11)., In addition a few cells lacked a second chromosome 21 and carried an additional der(21) t(?;21) (?;q22). The Vh segment in this cell line was Vh4-34 (Vh 4.21).
Derivation:

GA-10 was derived in 1996 from lymphoma cells circulating in the peripheral blood of a patient with a clinically aggressive, chemo-refractory Burkitts Lymphoma.

The patient had been treated unsuccessfully with cyclophosphamide, hydroxydaunomycin, vincristine, and prednisone (CHOP chemotherapy).

Comments:

The cells express relatively high levels of CD77 (approximately 90%); CD77 is also known as Burkitts lymphoma antigen (BLA).

GA-10 cells exhibit sensitivity to Shiga Toxin (Stx) in toxicity studies.

This cell line is slightly more sensitive to doxorubicin and etoposide than HL-60 leukemia cells.

Flavopiridol, a potent inhibitor of cell cycle progression through G1 and G2, produces apoptosis of the GA-10 cells at relatively low concentrations of 20-30 nM.

GA-10 cells are negative for Epstein-Barr virus (EBV).

Complete Growth Medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, ATCC 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (ATCC 30-2020) to a final concentration of 10%.
Subculturing: Cultures can be maintained by addition of fresh Medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 5 x 105 viable cells/mL. Maintain cultures at a cell concentration between 5 x 105 and 2 x 106 cells/mL. Do not allow the cell concentration to exceed 2 x 106 cells/mL.
Medium Renewal: Every 2 to 3 days

Complete culture medium 95%; DMSO, 5%. Cell culture tested DMSO is available as ATCC Catalog No. 4-X.
