| Comments： | Restriction digests of the clone give the following sizes (kb): HindIII/NotI--2.9, 0.78; XbaI--2.9, 0.50, 0.28; HindIII--3.7. The insert contains the following restriction sites (approximate kb from the 5 end): XbaI--0.49; StyI--0.40; SacI--0.68. The sequence surrounding the initiation codon was modified to 5-ACCAUGG-3 (Kozak consensus) to optimize translation. Thus the sequence between the vector insertion site and the initiating ATG is not derived from the mRNA. The p35 cDNA was cloned into pBluescript SK+ via HindIII to BamHI (Aug to stop), without recreating the BamHI site. |
