Medium : 45% Hams F10 + 45% Dulbeccos MEM + 10% h.i. FBS + 5 µg/ml human insulin + 5 µg/ml human transferrin + 50 nM hydrocortisone + 5 ng/ml selenite + 10 µM HEPES
Subculture : split confluent culture 1:4 every 6-7 days using trypsin/EDTA
Incubation : at 37 °C with 5% CO2
Doubling time : about 60-75 hours
Harvest : harvest of 2-3 x 106 cells/80 cm2
