Cell type : prostate carcinoma
Origin : cell line was derived from a human prostate carcinoma xenograft (CWR22R) that was serially propagated in nude mice after castration-induced regression and relapse of the parental, androgen-dependent CWR22 xenograft; the cells contain proviruses of the gammaretrovirus XMRV and this cell line has been assigned by the ZKBS (German Central Commission for Biological Safety) to risk category 2

Risk assessment : The cell line was shown to carry more than 10 copies of the xenotropic murine leukemia virus related virus integrated as proviruses into the eukaryotic genome. Active viruses are secreted into the cell culture supernatant. Murine retroviruses can cause leukemia, lymphoma or other tumors. A relation to analogous diseases in human could not be shown. Thus, the viruses and the cell line was categorized bio safety level 1. However, the German Commission for Biosafety (ZKBS) classified this cell line risk group 2 when the cells are used for genetic manipulations.

Medium : 40% RPMI 1640 + 40% Dulbeccos MEM + 20% h.i. FBS
Subculture : seed out at ca. 1-2 x 106 cells in a 175 cm2 flask; split confluent culture 1:3 to 1:10 every 3-6 days using trypsin/EDTA for ca. 10 min
Incubation : at 37 °C with 5% CO2
Doubling time : ca. 40-60 hours
Harvest : cell harvest of ca. 40 x 106 cells/175cm 2
Storage : frozen with 70% medium, 20% FBS, 10% DMSO
Mycoplasma : negative in DAPI, microbiological culture, RNA hybridization, PCR assays
